In washed human platelets the inhibition of platelet aggregation caused by serotonin or ristocetin is accompanied by an increase in the release of [14C]serotonin. The serotonin-induced release of [14C]serotonin is partially prevented by the platelet-specific phospholipase C inhibitor, quercetin-3-O-methyl ether. Since quercetin-3-O-methyl ether does not affect the serotonin autoreceptor, the present study was designed to determine if this autoreceptor could be functionally coupled to phospholipase C. It was found that quercetin-3-O-methyl ether inhibited serotonin-stimulated (125I)calmodulin binding in permeabilized platelets by 23%, whereas it had no effect on (125I)calmodulin binding to unstimulated platelets or to platelets activated by thrombin. The present data strongly suggest that the serotonin autoreceptor is functionally coupled to phosphatidylinositol-specific phospholipase C.
